The song tells the story of a woman who faces Jolene, a beautiful beautiful woman, whom she believes attempts to steal her lover and begs her "please do not take my son." Throughout the song, the woman begged Jolene "please do not take him just because you can." In 2012 a woman posted on the internet that she might be Jolene in the song. She says that she has met Dolly when she was at a concert when she was 7 years old.

According to Parton, the song was inspired by a red-headed banker who flirted with her husband Carl Dean at his local bank branch around when they were just married. In an interview, he also revealed that the name and appearance of Jolene is based on a young fan who came to the stage for his autograph.

This song became the number one solo song number one on the country charts after being released as a single at the end of 1973 (prior to the release of the album). Achieved the top spot in February 1974; it's also a moderate pop hit for her and contemporary little contemporary entries. In November 2016, the song has sold 733,000 digital copies in the US since it is available for digital download.

The song was released as a single later in the UK, and became the country's first top ten hit, reaching number seven on the UK Singles Chart in 1976. The song also re-entered the charts when Parton performed at the Glastonbury festival in 2014. The song has sold 255,300 digital copies in the UK in January 2017.

The single "Caroline" from Kirsty Maccoll in 1994 was an in-spirit response song for "Jolene," in which Jolene's character grieved at the loss of her best friend (titular Caroline) after having an affair with her husband.

In 2011, Estonia's indie rock band Ewert and Two Dragons released an answer song, also titled "Jolene," in response to the original recording. In this version, "man" Parton directly discuss Jolene, telling him that nothing will happen between them and that his current lover will fight to get him back.

In 2012, Canadian indie rock band, Hey Ocean! released a song titled "Jolene." The song borrowed from Parton's by singing part of the choir and then ignoring it by saying "But it's nothing but his favorite country song."

In 2013, country singer Jennifer Nettles recorded "The Girl," which she declared in the interview to be given the subtitle "The Ballad of Jolene." The song is written from the perspective of Jolene's character, which Nettle feels is unfairly slandered in the original song. In this version, another woman proves to be uninterested in taking another female gentleman, and the song is actually framed as a warning to the Parton character that her "husband" has a spinning eye. Dolly Parton's concept of paranoia about Jolene is also discussed in S01 E03 podcast musical comedy JimBob Music Massacre in February 2018.

In 2013, The Beautiful South guitarist David Rotheray recorded an album of answer tracks, among them "Jolene's Song," performed by Julie Murphy. This Jolene version is equally sad to know that the man who loves her belongs to another woman and expresses her pain as being painted as a heartless teaser by the original song.

In 2017, American singer-songwriter Cam released his single single in response to Parton's song. The song is sung from the standpoint of Jolene, where she sings for 'Diane', the character of Parton, and states that she does not know that her 'husband' is her husband. Cam noted to Rolling Stone Country that his song was "his response to Dolly Parton's' Jolene." That's an apology that so many couples deserve, but never got. Another woman came forward to deliver the news to his wife about the affair, respecting her enough to have such a difficult conversation, once she realized she was married. Because everyone should be able to decide their own way in life, based on the truth. Women mainly have to do this for each other, because our self-esteem can still be so wrapped up in our partners. And in true country mode, I have set the whole raw story for cheerful music, so you can dance while processing it all. "
